Overview

Zeiss LSM 880 upright confocal multiphoton microscope with Airyscan detector.

The LSM880 is a point scanning confocal microscope built on an inverted microscope body. There are a range of objectives, optimally chosen for in vivo or high resolution multiphoton microscopy. The microscope is all automated except for the condenser. The focus drive can be driven by the software, and a fast piezo drive is available (for z stacks up to 400 nm). Optionally, the system can be inverted to allow using the microscope as an upright imaging system using an LSMTech Inverterscope.

For confocal imaging, there are 6 continuous wave (CW) excitation laser lines and 2 pulsed lasers, one fixed at 1040 nm plus one variable (680-1300 nm) for Non Linear Optical (NLO)/ multiphoton microscopy.

For detection, a choice of 4 internal detectors: 2 photomultiplier tubes (PMTs) and 1 GaAsP detector in the scanhead using a shared pinhole, plus an Airyscan 32x GaAsP array. The Airyscan module can be used in several modes, enabling super-resolution, virtual pinhole or Airyscan fast modality. For multiphoton imaging, there is also a linear array of 4 external non descanned detectors (NDDs), 2 PMTs and 2 GaAsPs with a choice of filters in reflective position.

The system is equipped with a blacked out heated incubator and CO2 module to enable stable operating temperature for in vivo imaging.

Imaging Modes:

  • Confocal, Airyscan or Airyscan Fast fluorescence
  • 2 photon and 3 photon fluorescence
  • Second Harmonic Generation

All modalities can be performed in 2D (xy) or 3D (xyz), multichannel with timelapse and/or tiling and/or multiple locations, plus ROI scanning allowing FRAP and FRET.
