Overview
The LSM700 is a point scanning confocal microscope built on an inverted microscope body.
There are a full range of objectives and the microscope is automated except for the condenser arm and stage. The focus drive can be driven by the software, making z stacks possible.
For confocal imaging, there are 3 excitation laser lines (405, 488 and 561 nm) and for detection, 2 photomultiplier tubes (PMTs) in the scanhead using a shared pinhole which can be separated by a sliding dichroic, plus a transmission PMT for brightfield.
2 or 3 colour fluorescence imaging is therefore easy, and 4 or 5 colour imaging too, provided that good separation of the fluorophores is possible.
Finally, for short term live cell imaging, a stage top incubator is available, with temperature and CO2 regulation.
Imaging modes:
- Spot scanning, line scanning, 2D (xy) or 3D (xyz)
- multichannel with timelapse,
- ROI scanning with bleaching, allowing FRAP
