Clinical interests
I am a practicing clinician, with regular clinics in adult infectious diseases and a joint clinic with colleagues in Immunology. My specialist interests include severe and/or high consequence viral disease, immunodeficiency, and autoinflammatory disease.
Research Interests
Discovery research
Our research aims to define protective and deleterious mechanisms of innate antiviral immunity, in the anticipation that such knowledge may reveal new therapeutic opportunities in viral and inflammatory disease. The processes by which cells detect viral pathogens are reasonably well understood, however less is known about how the downstream response(s) contribute to disease outcome. In this respect, inborn errors of immunity provide a rich resource, teaching us unique lessons about the action and regulation of antiviral signalling in humans.
Type I interferons (IFN-I) are central to the innate response to virus infection. Previously, we have identified human inborn errors which either impair or enhance IFN-I receptor signalling, leading respectively to viral disease or sterile autoinflammation. Thus precise calibration of this response appears essential to host defence and immune homeostasis.
We are interested in the cellular and molecular regulation of IFN-I signalling and how this contributes to antiviral defence in tissues. We employ cutting-edge techniques of disease modelling, combining CRISPR/Cas9 gene editing in human induced pluripotent stem cell systems with studies in model organisms. We work closely with Professor Sophie Hambleton's Primary Immunodeficiency Group: http://www.ncl.ac.uk/icm/people/profile/sophie.hambleton and a range of national and international collaborators.
Current projects
Building on a longstanding interest in antiviral responses of macrophages, a current focus is on microglia, the resident macrophages of the brain parenchyma, and the role these cells play in responding to neurotropic viruses and coordinating the brain's IFN network.
Another area of focus is how innate IFNs intersect in antiviral defence of the airway epithelium.
Finally, we are interested in modelling sterile IFN-I mediated pathology using in vivo models of human disease entities known as type I interferonopathies.
Recently, in response to the SARS-CoV-2 pandemic, we have embarked on a programme of basic and translational research to characterise immune responses to SARS-CoV-2, through involvement in the UK Coronavirus Immunology Consortium (UK-CIC) and the PITCH consortium.
Clinical ID Research
Working with partners in Newcastle upon Tyne Hospitals NHS Foundation Trust and the NIHR North East and North Cumbria Clinical Research Network, I lead the delivery of a diverse clinical research portfolio of phase I-III vaccine and interventional studies in Infectious Disease.
